VACUNA PARA VIRUS EBOLA ZAIRE (ERVEBO®)4, 45,46
El principio activo de Ervebo® es un virus vivo atenuado recombinante
de la estomatitis vesicular, con la eliminación de la
glicoproteína de la envoltura de la estomatitis vesicular, reemplazada
por la glicoproteína de superficie del Ébola virus Zaire. Esta
vacuna proporciona una inmunización activa contra el virus del
Ébola Zaire mediante la inducción de una respuesta inmunitaria
que ayuda a proteger contra la enfermedad. El Comité Europeo
de Evaluación de Medicamentos ha recomendado de manera
condicional la autorización de comercialización al satisfacer una
necesidad médica no cubierta, en la medida en que, el beneficio
para la salud pública de su inmediata disponibilidad es superior
al riesgo inherente de que todavía se requieran datos adicionales.
En los ensayos clínicos ha mostrado prevenir la enfermedad
por el virus del Ébola confirmada en laboratorio, basándose en
los datos de eficacia clínica recopilados en individuos con riesgo
de infección durante un brote de Ébola en África occidental.
Los efectos adversos más frecuentes fueron reacciones en
el lugar de inyección (dolor, inflamación y eritema), cefalea,
pirexia, mialgia, fatiga y artralgia. En un ensayo fase 3, doble
ciego controlado con placebo en voluntarios sanos se estudiaron
acontecimientos adversos (a nivel local y regional) presentados
del día 1 al 42 tras la vacunación, ampliándose el tiempo de seguimiento
a 6 meses en los acontecimientos adversos clasificados
como graves. Fiebre se observó en el 20,2% de los participantes,
siguiendo en frecuencia: artralgias (17,1%), artritis (5,1%) y rash
(3,8%). Se notificaron 21 acontecimientos adversos graves y 2
muertes, que fueron valorados por los investigadores como no
relacionados con la vacuna. Los autores concluyen que la vacuna
fue en general bien tolerada, y que apoya su uso en personas
de alto riesgo de enfermedad.
